DESCRIPTION

Targeting cancer complexity at origin and progression

Our research is focused on better understanding breast cancer initiation and, from acquired knowledge, to assess targeted prevention strategies. In parallel, we study pan-cancer vulnerabilities and the connection to a rare, lung metastatic disease, lymphangioleiomyomatosis (LAM).

Polygenic risk scores and breast and epithelial ovarian cancer risks for carriers of BRCA1 and BRCA2 pathogenic variants

    16 de July de 2020/in Pujana's Lab/by miguel

Genet Med. 2020 Oct;22(10):1653-1666. doi: 10.1038/s41436-020-0862-x. Epub 2020 Jul 15. ABSTRACT PURPOSE: We assessed the associations between population-based polygenic risk scores (PRS) for breast (BC) or epithelial ovarian cancer (EOC) with cancer risks for BRCA1 and BRCA2 pathogenic variant carriers. METHODS: Retrospective cohort data on 18,935 BRCA1 and 12,339 BRCA2 female pathogenic variant carriers of […]

Immune Cell Associations with Cancer Risk

    5 de July de 2020/in Pujana's Lab/by miguel

iScience. 2020 Jul 24;23(7):101296. doi: 10.1016/j.isci.2020.101296. Epub 2020 Jun 20. ABSTRACT Proper immune system function hinders cancer development, but little is known about whether genetic variants linked to cancer risk alter immune cells. Here, we report 57 cancer risk loci associated with differences in immune and/or stromal cell contents in the corresponding tissue. Predicted target […]

Characterization of the Cancer Spectrum in Men With Germline BRCA1 and BRCA2 Pathogenic Variants: Results From the Consortium of Investigators of Modifiers of BRCA1/2 (CIMBA)

    3 de July de 2020/in Pujana's Lab/by miguel

JAMA Oncol. 2020 Aug 1;6(8):1218-1230. doi: 10.1001/jamaoncol.2020.2134. ABSTRACT IMPORTANCE: The limited data on cancer phenotypes in men with germline BRCA1 and BRCA2 pathogenic variants (PVs) have hampered the development of evidence-based recommendations for early cancer detection and risk reduction in this population. OBJECTIVE: To compare the cancer spectrum and frequencies between male BRCA1 and BRCA2 […]

NEK10 tyrosine phosphorylates p53 and controls its transcriptional activity

    21 de June de 2020/in Pujana's Lab/by miguel

Oncogene. 2020 Jul;39(30):5252-5266. doi: 10.1038/s41388-020-1361-x. Epub 2020 Jun 19. ABSTRACT In response to genotoxic stress, multiple kinase signaling cascades are activated, many of them directed towards the tumor suppressor p53, which coordinates the DNA damage response (DDR). Defects in DDR pathways lead to an accumulation of mutations that can promote tumorigenesis. Emerging evidence implicates multiple […]

Tumors defective in homologous recombination rely on oxidative metabolism: relevance to treatments with PARP inhibitors

    14 de May de 2020/in Pujana's Lab/by miguel

EMBO Mol Med. 2020 Jun 8;12(6):e11217. doi: 10.15252/emmm.201911217. Epub 2020 May 13. ABSTRACT Mitochondrial metabolism and the generation of reactive oxygen species (ROS) contribute to the acquisition of DNA mutations and genomic instability in cancer. How genomic instability influences the metabolic capacity of cancer cells is nevertheless poorly understood. Here, we show that homologous recombination-defective […]

    Research Projects

The group works and collaborates to decipher the systems mechanisms of cancer development and progression, with the aim of developing more precise preventive and therapeutic approaches. The group combines in silico and wet-lab studies for the purpose of identifying cancer principles and vulnerabilities.
