DESCRIPTION

Targeting cancer complexity at origin and progression

Our research is focused on better understanding breast cancer initiation and, from acquired knowledge, to assess targeted prevention strategies. In parallel, we study pan-cancer vulnerabilities and the connection to a rare, lung metastatic disease, lymphangioleiomyomatosis (LAM).

Long-term results of sirolimus treatment in lymphangioleiomyomatosis: a single referral centre experience

    14 de May de 2021/in Pujana's Lab/by miguel

Sci Rep. 2021 May 13;11(1):10171. doi: 10.1038/s41598-021-89562-0. ABSTRACT There are few published data on long-term treatment with sirolimus in lymphangioleiomyomatosis (LAM). The objective of this study was to describe the long-term effect of sirolimus in a series of LAM patients followed up in a referral centre, focusing on pulmonary function. We retrospectively reviewed a series […]

A case-only study to identify genetic modifiers of breast cancer risk for BRCA1/BRCA2 mutation carriers

Nat Commun. 2021 Feb 17;12(1):1078. doi: 10.1038/s41467-020-20496-3. ABSTRACT Breast cancer (BC) risk for BRCA1 and BRCA2 mutation carriers varies by genetic and familial factors. About 50 common variants have been shown to modify BC risk for mutation carriers. All but three, were identified in general population studies. Other mutation carrier-specific susceptibility variants may exist but […]

Loss of TGFβ signaling increases alternative end-joining DNA repair that sensitizes to genotoxic therapies across cancer types

    11 de February de 2021/in Pujana's Lab/by miguel

Sci Transl Med. 2021 Feb 10;13(580):eabc4465. doi: 10.1126/scitranslmed.abc4465. ABSTRACT Among the pleotropic roles of transforming growth factor-β (TGFβ) signaling in cancer, its impact on genomic stability is least understood. Inhibition of TGFβ signaling increases use of alternative end joining (alt-EJ), an error-prone DNA repair process that typically functions as a “backup” pathway if double-strand break […]

RNF168 regulates R-loop resolution and genomic stability in BRCA1/2-deficient tumors

    2 de February de 2021/in Pujana's Lab/by miguel

J Clin Invest. 2021 Feb 1;131(3):e140105. doi: 10.1172/JCI140105. ABSTRACT Germline mutations in BRCA1 and BRCA2 (BRCA1/2) genes considerably increase breast and ovarian cancer risk. Given that tumors with these mutations have elevated genomic instability, they exhibit relative vulnerability to certain chemotherapies and targeted treatments based on poly (ADP-ribose) polymerase (PARP) inhibition. However, the molecular mechanisms […]

    Research Projects

The group works and collaborates to decipher the systems mechanisms of cancer development and progression, with the aim of developing more precise preventive and therapeutic approaches. The group combines in silico and wet-lab studies for the purpose of identifying cancer principles and vulnerabilities.
