Metformin directly targets the H3K27me3 demethylase KDM6A/UTX

10 de May de 2018/in Menendez's Lab/by miguel

Aging Cell. 2018 Aug;17(4):e12772. doi: 10.1111/acel.12772. Epub 2018 May 8. ABSTRACT Metformin, the first drug chosen to be tested in a clinical trial aimed to target the biology of aging per se, has been clinically exploited for decades in the absence of a complete understanding of its therapeutic targets or chemical determinants. We here outline […]

Silibinin is a direct inhibitor of STAT3

17 de April de 2018/in Menendez's Lab/by miguel

Food Chem Toxicol. 2018 Jun;116(Pt B):161-172. doi: 10.1016/j.fct.2018.04.028. Epub 2018 Apr 14. ABSTRACT We herein combined experimental and computational efforts to delineate the mechanism of action through which the flavonolignan silibinin targets STAT3. Silibinin reduced IL-6 inducible, constitutive, and acquired feedback activation of STAT3 at tyrosine 705 (Y705). Silibinin attenuated the inducible phospho-activation of Y705 […]

Assessment of extracellular matrix-related biomarkers in patients with lower extremity artery disease

5 de April de 2018/in Menendez's Lab/by miguel

J Vasc Surg. 2018 Oct;68(4):1135-1142.e6. doi: 10.1016/j.jvs.2017.12.071. Epub 2018 Mar 31. ABSTRACT BACKGROUND: The prevalence of lower extremity artery disease (LEAD) is high (20%-25%) in the population older than 65 years, but patients are seldom identified until the disease is advanced. Circulating markers of disease activity might provide patients with a key opportunity for timely […]

Epigenetic regulation of cell fate reprogramming in aging and disease: A predictive computational model

16 de March de 2018/in Menendez's Lab/by miguel

PLoS Comput Biol. 2018 Mar 15;14(3):e1006052. doi: 10.1371/journal.pcbi.1006052. eCollection 2018 Mar. ABSTRACT Understanding the control of epigenetic regulation is key to explain and modify the aging process. Because histone-modifying enzymes are sensitive to shifts in availability of cofactors (e.g. metabolites), cellular epigenetic states may be tied to changing conditions associated with cofactor variability. The aim […]

Extra-virgin olive oil contains a metabolo-epigenetic inhibitor of cancer stem cells

17 de February de 2018/in Menendez's Lab/by miguel

Carcinogenesis. 2018 Apr 5;39(4):601-613. doi: 10.1093/carcin/bgy023. ABSTRACT Targeting tumor-initiating, drug-resistant populations of cancer stem cells (CSC) with phytochemicals is a novel paradigm for cancer prevention and treatment. We herein employed a phenotypic drug discovery approach coupled to mechanism-of-action profiling and target deconvolution to identify phenolic components of extra virgin olive oil (EVOO) capable of suppressing […]
