Metabolism and cancer
Menendez’s Lab

DESCRIPTION

Unlocking the metabolic secrets of cancer: Our “Metabolism and Cancer” team studies how cellular metabolism drives cancer growth, drug resistance, and metastasis. We use cutting-edge laboratory experiments, computational tools and clinical trials to validate our discoveries in cancer patients.

A phase 2 trial of neoadjuvant metformin in combination with trastuzumab and chemotherapy in women with early HER2-positive breast cancer: the METTEN study

28 de November de 2018/in Menendez's Lab/by miguel

Oncotarget. 2018 Nov 2;9(86):35687-35704. doi: 10.18632/oncotarget.26286. eCollection 2018 Nov 2. ABSTRACT The METTEN study assessed the efficacy, tolerability, and safety of adding metformin to neoadjuvant chemotherapy plus trastuzumab in early HER2-positive breast cancer (BC). Women with primary, non-metastatic HER2-positive BC were randomized (1:1) to receive metformin (850 mg twice-daily) for 24 weeks concurrently with 12 […]

Metformin Is a Direct SIRT1-Activating Compound: Computational Modeling and Experimental Validation

22 de November de 2018/in Menendez's Lab/by miguel

Front Endocrinol (Lausanne). 2018 Nov 6;9:657. doi: 10.3389/fendo.2018.00657. eCollection 2018. ABSTRACT Metformin has been proposed to operate as an agonist of SIRT1, a nicotinamide adenine dinucleotide (NAD+)-dependent deacetylase that mimics most of the metabolic responses to calorie restriction. Herein, we present an in silico analysis focusing on the molecular docking and dynamic simulation of the […]

An olive oil phenolic is a new chemotype of mutant isocitrate dehydrogenase 1 (IDH1) inhibitors

15 de November de 2018/in Menendez's Lab/by miguel

Carcinogenesis. 2019 Mar 12;40(1):27-40. doi: 10.1093/carcin/bgy159. ABSTRACT Mutations in the isocitrate dehydrogenase 1 (IDH1) gene confer an oncogenic gain-of-function activity that allows the conversion of α-ketoglutarate (α-KG) to the oncometabolite R-2-hydroxyglutarate (2HG). The accumulation of 2HG inhibits α-KG-dependent histone and DNA demethylases, thereby generating genome-wide hypermethylation phenotypes with cancer-initiating properties. Several chemotypes of mutant IDH1/2-targeted […]

Influence of age and spectral power distribution on mesopic visual sensitivity

1 de November de 2018/in Menendez's Lab/by miguel

Atten Percept Psychophys. 2019 Feb;81(2):504-516. doi: 10.3758/s13414-018-1616-6. ABSTRACT The ability of the human visual system to detect stimuli at low illumination levels provides awareness of potential risks. The influence of age and spectral power distribution on mesopic spectral sensitivity is analyzed. Two typical light sources are used, a high-pressure sodium lamp (HPS), with a higher […]

Plasma Energy-Balance Metabolites Discriminate Asymptomatic Patients with Peripheral Artery Disease

18 de October de 2018/in Menendez's Lab/by miguel

Mediators Inflamm. 2018 Sep 20;2018:2760272. doi: 10.1155/2018/2760272. eCollection 2018. ABSTRACT Peripheral artery disease (PAD) is a common disease affecting 20-25% of population over 60 years old. Early diagnosis is difficult because symptoms only become evident in advanced stages of the disease. Inflammation, impaired metabolism, and mitochondrial dysfunction predispose to PAD, which is normally associated with […]
