Therapeutic resistance and hematological tumors
Sureda’s & Farre’s Lab

DESCRIPTION

Our group aims to gain a better understanding of the clinical characteristics and mechanisms of progression and therapeutic resistance of hematological tumors with a focus on multiple myeloma and aggressive T and B lymphomas to propose new therapeutic strategies.

The lab is interested in improving the understanding of the molecular mechanisms leading to the progression and therapeutic resistance of multiple myeloma, including extramedullary disease, Hodgkin lymphoma and aggressive non-Hodgkin B and T lymphomas.

To develop these lines of research, we use patient samples and advanced preclinical models for therapeutic, mechanistic and functional studies. We have developed the technology to generate murine models derived from patients by orthotopic implantation (orthotopic PDX) and 2D/3D cultures that we can humanize using components of the human immune system (from the same patient from whom the model has been derived, when possible). Additionally, we are involved in a project that is developing humanized nanomedicines as a new therapeutic strategy for acute myeloid leukemia.

Our group is integrated by professionals from the Clinical Hematology Service of ICO/Hospitalet, the ProCURE/ICO Program and the Pathology Service of the Hospital of Bellvitge. Within the Hematology Service of the ICO Hospitalet, one of our main objectives is to promote translational research training for the professionals of the Service, generating opportunities for the completion of doctoral theses and stimulating new scientific leadership as well as to attract talent with a mixed clinical/researcher profile for the Service.

Decreased RORC expression and downstream signaling in HTLV-1-associated adult T-cell lymphoma/leukemia uncovers an antiproliferative IL17 link: A potential target for immunotherapy?

11 de October de 2018/in Sureda's & Farre's Lab/by miguel

Int J Cancer. 2019 Apr 1;144(7):1664-1675. doi: 10.1002/ijc.31922. Epub 2018 Dec 18. ABSTRACT Retinoic acid-related drugs have shown promising pre-clinical activity in Adult T-cell Leukemia/Lymphoma, but RORC signaling has not been explored. Therefore, we investigated transcriptome-wide interactions of the RORC pathway in HTLV-1 and ATL, using our own and publicly available gene expression data for […]

Early Juvenile Human T-cell Lymphotropic Virus Type-1-Associated Myelopathy/Tropical Spastic Paraparesis: Study of 25 Patients

19 de June de 2018/in Sureda's & Farre's Lab/by miguel

Clin Infect Dis. 2018 Oct 15;67(9):1427-1433. doi: 10.1093/cid/ciy289. ABSTRACT BACKGROUND: Human T-cell lymphotropic virus type-1 (HTLV-1) may cause severe diseases such as HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP) and infective dermatitis associated with HTLV-1 (IDH). The clinical characteristics and progression of 25 early onset HAM/TSP associated or not to IDH were described. METHODS: Following-up 37 IDH […]

A genetic IFN/STAT1/FAS axis determines CD4 T stem cell memory levels and apoptosis in healthy controls and Adult T-cell Leukemia patients

4 de May de 2018/in Sureda's & Farre's Lab/by miguel

Oncoimmunology. 2018 Feb 13;7(5):e1426423. doi: 10.1080/2162402X.2018.1426423. eCollection 2018. ABSTRACT Adult T-cell leukemia (ATL) is an aggressive, chemotherapy-resistant CD4+CD25+ leukemia caused by HTLV-1 infection, which usually develops in a minority of patients several decades after infection. IFN + AZT combination therapy has shown clinical benefit in ATL, although its mechanism of action remains unclear. We have […]

Tumor xenograft modeling identifies an association between TCF4 loss and breast cancer chemoresistance

19 de April de 2018/in Sureda's & Farre's Lab/by miguel

Dis Model Mech. 2018 May 18;11(5):dmm032292. doi: 10.1242/dmm.032292. ABSTRACT Understanding the mechanisms of cancer therapeutic resistance is fundamental to improving cancer care. There is clear benefit from chemotherapy in different breast cancer settings; however, knowledge of the mutations and genes that mediate resistance is incomplete. In this study, by modeling chemoresistance in patient-derived xenografts (PDXs), […]
