DESCRIPTION

Targeting cancer complexity at origin and progression

Our research is focused on better understanding breast cancer initiation and, from acquired knowledge, to assess targeted prevention strategies. In parallel, we study pan-cancer vulnerabilities and the connection to a rare, lung metastatic disease, lymphangioleiomyomatosis (LAM).

Biological basis of extensive pleiotropy between blood traits and cancer risk

    3 de February de 2024/in Pujana's Lab/by

Genome Med. 2024 Feb 2;16(1):21. doi: 10.1186/s13073-024-01294-8. ABSTRACT BACKGROUND: The immune system has a central role in preventing carcinogenesis. Alteration of systemic immune cell levels may increase cancer risk. However, the extent to which common genetic variation influences blood traits and cancer risk remains largely undetermined. Here, we identify pleiotropic variants and predict their underlying […]

Topoisomerase 1 Inhibition in MYC-Driven Cancer Promotes Aberrant R-Loop Accumulation to Induce Synthetic Lethality

    21 de November de 2023/in Pujana's Lab/by

Cancer Res. 2023 Nov 21:OF1-OF15. doi: 10.1158/0008-5472.CAN-22-2948. Online ahead of print. ABSTRACT MYC is a central regulator of gene transcription and is frequently dysregulated in human cancers. As targeting MYC directly is challenging, an alternative strategy is to identify specific proteins or processes required for MYC to function as a potent cancer driver that can […]

Differential DNA damage repair and PARP inhibitor vulnerability of the mammary epithelial lineages

    17 de October de 2023/in Pujana's Lab/by miguel

Cell Rep. 2023 Oct 31;42(10):113256. doi: 10.1016/j.celrep.2023.113256. Epub 2023 Oct 16. ABSTRACT It is widely assumed that all normal somatic cells can equally perform homologous recombination (HR) and non-homologous end joining in the DNA damage response (DDR). Here, we show that the DDR in normal mammary gland inherently depends on the epithelial cell lineage identity. […]

RNF8 ubiquitylation of XRN2 facilitates R-loop resolution and restrains genomic instability in BRCA1 mutant cells

    12 de September de 2023/in Pujana's Lab/by miguel

Nucleic Acids Res. 2023 Oct 27;51(19):10484-10505. doi: 10.1093/nar/gkad733. ABSTRACT Breast cancer linked with BRCA1/2 mutations commonly recur and resist current therapies, including PARP inhibitors. Given the lack of effective targeted therapies for BRCA1-mutant cancers, we sought to identify novel targets to selectively kill these cancers. Here, we report that loss of RNF8 significantly protects Brca1-mutant […]

    Research Projects

The group works and collaborates to decipher the systems mechanisms of cancer development and progression, with the aim of developing more precise preventive and therapeutic approaches. The group combines in silico and wet-lab studies for the purpose of identifying cancer principles and vulnerabilities.
