DESCRIPTION

Targeting cancer complexity at origin and progression

Our research is focused on better understanding breast cancer initiation and, from acquired knowledge, to assess targeted prevention strategies. In parallel, we study pan-cancer vulnerabilities and the connection to a rare, lung metastatic disease, lymphangioleiomyomatosis (LAM).

Cancer genetic counseling in Chile: Addressing barriers, confronting challenges, and seizing opportunities in an underserved Latin American Community

    23 de December de 2024/in Pujana's Lab/by

Genet Med Open. 2024 Oct 30;2(Suppl 2):101898. doi: 10.1016/j.gimo.2024.101898. eCollection 2024. ABSTRACT PURPOSE: Despite the rapid advancements in genomics and the enactment of a new cancer law in Chile, the implementation of cancer genetic counseling continues to face significant challenges because of limited resources and infrastructure. METHODS: We conducted a survey targeting health care providers […]

Fatty acid synthase (FASN) is a tumor-cell-intrinsic metabolic checkpoint restricting T-cell immunity

    30 de September de 2024/in Pujana's Lab/by

Cell Death Discov. 2024 Sep 30;10(1):417. doi: 10.1038/s41420-024-02184-z. ABSTRACT Fatty acid synthase (FASN)-catalyzed endogenous lipogenesis is a hallmark of cancer metabolism. However, whether FASN is an intrinsic mechanism of tumor cell defense against T cell immunity remains unexplored. To test this hypothesis, here we combined bioinformatic analysis of the FASN-related immune cell landscape, real-time assessment […]

DNA double-strand break-capturing nuclear envelope tubules drive DNA repair

    17 de April de 2024/in Pujana's Lab/by

Nat Struct Mol Biol. 2024 Apr 17. doi: 10.1038/s41594-024-01286-7. Online ahead of print. ABSTRACT Current models suggest that DNA double-strand breaks (DSBs) can move to the nuclear periphery for repair. It is unclear to what extent human DSBs display such repositioning. Here we show that the human nuclear envelope localizes to DSBs in a manner […]

Large-scale genome-wide association study of 398,238 women unveils seven novel loci associated with high-grade serous epithelial ovarian cancer risk

    18 de March de 2024/in Pujana's Lab/by

medRxiv [Preprint]. 2024 Mar 4:2024.02.29.24303243. doi: 10.1101/2024.02.29.24303243. ABSTRACT BACKGROUND: Nineteen genomic regions have been associated with high-grade serous ovarian cancer (HGSOC). We used data from the Ovarian Cancer Association Consortium (OCAC), Consortium of Investigators of Modifiers of BRCA1 / BRCA2 (CIMBA), UK Biobank (UKBB), and FinnGen to identify novel HGSOC susceptibility loci and develop polygenic […]

Identification of differential biological activity and synergy between the PARP inhibitor rucaparib and its major metabolite

    9 de February de 2024/in Pujana's Lab/by

Cell Chem Biol. 2024 Feb 6:S2451-9456(24)00043-6. doi: 10.1016/j.chembiol.2024.01.007. Online ahead of print. ABSTRACT The (poly)pharmacology of drug metabolites is seldom comprehensively characterized in drug discovery. However, some drug metabolites can reach high plasma concentrations and display in vivo activity. Here, we use computational and experimental methods to comprehensively characterize the kinase polypharmacology of M324, the […]
