Cancer immunotherapy
Alemany’s & Piulats’s lab

DESCRIPTION

Our main aim is to develop new anticancer treatments based on novel designs of oncolytic adenoviruses, combining them with mesenchymal stem cells and cellular therapies derived from tumor reactive T-lymphocytes against solid tumors.

The Cancer Immunotherapy group, led by Dr Piulats and Dr Alemany, is dedicated to develop new immunotherapies based on oncolytic adenoviruses or advanced therapy medicinal products (ATMPs) to treat the so-called “cold” tumors and rare malignancies. Oncolytic viruses represent a double antitumor strategy: they have an oncolytic activity and on top of that, they promote lymphocyte infiltration and immune responses against tumors. We are designing and evaluating new oncolytic adenovirus to improve their Intratumor diffusion and tumor reactivity and testing those using murine models. We are developing CAR-NK cells models, cell therapies based on autoreactive TCRs from patients and combined ATMP using virus and mesenchymal stem cells. We combine clinical and translational research, and we have led different clinical trials based on immunotherapy, demonstrating the feasibility of immunotherapy approaches to treat this kind of tumors.

SMARCA4 deficient tumours are vulnerable to KDM6A/UTX and KDM6B/JMJD3 blockade

15 de July de 2021/in Alemany’s & Piulats’s lab/by

Nat Commun. 2021 Jul 14;12(1):4319. doi: 10.1038/s41467-021-24618-3. ABSTRACT Despite the genetic inactivation of SMARCA4, a core component of the SWI/SNF-complex commonly found in cancer, there are no therapies that effectively target SMARCA4-deficient tumours. Here, we show that, unlike the cells with activated MYC oncogene, cells with SMARCA4 inactivation are refractory to the histone deacetylase inhibitor, […]

Combination of chemotherapy with BRAF inhibitors results in effective eradication of malignant melanoma by preventing ATM-dependent DNA repair

18 de June de 2021/in Alemany’s & Piulats’s lab/by

Oncogene. 2021 Aug;40(31):5042-5048. doi: 10.1038/s41388-021-01879-2. Epub 2021 Jun 17. ABSTRACT Invasive malignant melanoma (MM) is an aggressive tumor with no curative therapy in advanced stages. Chemotherapy has not demonstrated its efficacy in MM and current treatment for tumors carrying the most frequent BRAFV600E mutation consists of BRAF inhibitors alone or in combination with MAPK pathway […]

KEYNOTE-921: Phase III study of pembrolizumab plus docetaxel for metastatic castration-resistant prostate cancer

8 de June de 2021/in Alemany’s & Piulats’s lab/by

Future Oncol. 2021 Sep;17(25):3291-3299. doi: 10.2217/fon-2020-1133. Epub 2021 Jun 8. ABSTRACT Despite recent advances, treatment options for men with metastatic castration-resistant prostate cancer (mCRPC) progressing after next-generation hormonal agents (NHAs) are limited and provide only modest survival benefit. Thus, an unmet need remains for mCRPC patients after treatment with targeted endocrine therapy or NHA therapy. […]

Predicting MHC I restricted T cell epitopes in mice with NAP-CNB, a novel online tool

25 de May de 2021/in Alemany’s & Piulats’s lab/by

Sci Rep. 2021 May 24;11(1):10780. doi: 10.1038/s41598-021-89927-5. ABSTRACT Lack of a dedicated integrated pipeline for neoantigen discovery in mice hinders cancer immunotherapy research. Novel sequential approaches through recurrent neural networks can improve the accuracy of T-cell epitope binding affinity predictions in mice, and a simplified variant selection process can reduce operational requirements. We have developed […]

Characterization of the Endometrial MSC Marker Ectonucleoside Triphosphate Diphosphohydrolase-2 (NTPDase2/CD39L1) in Low- and High-Grade Endometrial Carcinomas: Loss of Stromal Expression in the Invasive Phenotypes

30 de April de 2021/in Alemany’s & Piulats’s lab/by

J Pers Med. 2021 Apr 22;11(5):331. doi: 10.3390/jpm11050331. ABSTRACT Ectonucleoside triphosphate diphosphohydrolase-2 (NTPDase2/CD39L1) has been described in human non-pathological endometrium in both epithelial and stromal components without changes along the cycle. It was identified as a stromal marker of basalis. In the present study, we aimed to evaluate NTPDase2 distribution, using immunolabeling and in situ […]
