Cancer immunotherapy
Alemany’s & Piulats’s lab

DESCRIPTION

Our main aim is to develop new anticancer treatments based on novel designs of oncolytic adenoviruses, combining them with mesenchymal stem cells and cellular therapies derived from tumor reactive T-lymphocytes against solid tumors.

The Cancer Immunotherapy group, led by Dr Piulats and Dr Alemany, is dedicated to develop new immunotherapies based on oncolytic adenoviruses or advanced therapy medicinal products (ATMPs) to treat the so-called “cold” tumors and rare malignancies. Oncolytic viruses represent a double antitumor strategy: they have an oncolytic activity and on top of that, they promote lymphocyte infiltration and immune responses against tumors. We are designing and evaluating new oncolytic adenovirus to improve their Intratumor diffusion and tumor reactivity and testing those using murine models. We are developing CAR-NK cells models, cell therapies based on autoreactive TCRs from patients and combined ATMP using virus and mesenchymal stem cells. We combine clinical and translational research, and we have led different clinical trials based on immunotherapy, demonstrating the feasibility of immunotherapy approaches to treat this kind of tumors.

Health-Related Quality of Life of Patients with Recurrent or Metastatic Cutaneous Squamous Cell Carcinoma Treated with Pembrolizumab in KEYNOTE-629

24 de September de 2021/in Alemany’s & Piulats’s lab/by

Dermatol Ther (Heidelb). 2021 Oct;11(5):1777-1790. doi: 10.1007/s13555-021-00598-6. Epub 2021 Sep 23. ABSTRACT INTRODUCTION: Pembrolizumab provided durable responses and acceptable safety in recurrent or metastatic (R/M) cutaneous squamous cell carcinoma (cSCC) in the KEYNOTE-629 study. In this elderly, fragile population with disfiguring tumours, preservation of health-related quality of life (HRQoL) is critical. Here, we present pre-specified […]

Overall Survival Benefit with Tebentafusp in Metastatic Uveal Melanoma

22 de September de 2021/in Alemany’s & Piulats’s lab/by

N Engl J Med. 2021 Sep 23;385(13):1196-1206. doi: 10.1056/NEJMoa2103485. ABSTRACT BACKGROUND: Uveal melanoma is a disease that is distinct from cutaneous melanoma, with a low tumor mutational burden and a 1-year overall survival of approximately 50% in patients with metastatic uveal melanoma. Data showing a proven overall survival benefit with a systemic treatment are lacking. […]

A single-cell tumor immune atlas for precision oncology

22 de September de 2021/in Alemany’s & Piulats’s lab/by

Genome Res. 2021 Oct;31(10):1913-1926. doi: 10.1101/gr.273300.120. Epub 2021 Sep 21. ABSTRACT The tumor immune microenvironment is a main contributor to cancer progression and a promising therapeutic target for oncology. However, immune microenvironments vary profoundly between patients, and biomarkers for prognosis and treatment response lack precision. A comprehensive compendium of tumor immune cells is required to […]

Hyaluronidase expression within tumors increases virotherapy efficacy and T cell accumulation

11 de August de 2021/in Alemany’s & Piulats’s lab/by

Mol Ther Oncolytics. 2021 May 29;22:27-35. doi: 10.1016/j.omto.2021.05.009. eCollection 2021 Sep 24. ABSTRACT Oncolytic viruses (OVs) preferentially infect and selectively replicate in cancer cells. OVs have been tested in clinical trials as monotherapy or in combination with chemotherapy, radiotherapy, and immunotherapy. However, the dense extracellular matrix hampers the intratumoral spreading and efficacy of OVs. Previously […]

Transgene codon usage drives viral fitness and therapeutic efficacy in oncolytic adenoviruses

28 de July de 2021/in Alemany’s & Piulats’s lab/by

NAR Cancer. 2021 Apr 26;3(2):zcab015. doi: 10.1093/narcan/zcab015. eCollection 2021 Jun. ABSTRACT Arming oncolytic adenoviruses with therapeutic transgenes is a well-established strategy for multimodal tumour attack. However, this strategy sometimes leads to unexpected attenuated viral replication and a loss of oncolytic effects, preventing these viruses from reaching the clinic. Previous work has shown that altering codon […]
