Cancer immunotherapy
Alemany’s & Piulats’s lab

DESCRIPTION

Our main aim is to develop new anticancer treatments based on novel designs of oncolytic adenoviruses, combining them with mesenchymal stem cells and cellular therapies derived from tumor reactive T-lymphocytes against solid tumors.

The Cancer Immunotherapy group, led by Dr Piulats and Dr Alemany, is dedicated to develop new immunotherapies based on oncolytic adenoviruses or advanced therapy medicinal products (ATMPs) to treat the so-called “cold” tumors and rare malignancies. Oncolytic viruses represent a double antitumor strategy: they have an oncolytic activity and on top of that, they promote lymphocyte infiltration and immune responses against tumors. We are designing and evaluating new oncolytic adenovirus to improve their Intratumor diffusion and tumor reactivity and testing those using murine models. We are developing CAR-NK cells models, cell therapies based on autoreactive TCRs from patients and combined ATMP using virus and mesenchymal stem cells. We combine clinical and translational research, and we have led different clinical trials based on immunotherapy, demonstrating the feasibility of immunotherapy approaches to treat this kind of tumors.

Oncolytic adenovirus coding for shedding-resistant MICA enhances immune responses against tumors

5 de January de 2024/in Alemany’s & Piulats’s lab/by

Cancer Immunol Immunother. 2024 Jan 5;73(1):5. doi: 10.1007/s00262-023-03611-3. ABSTRACT Cancer immunotherapies strive to overcome tumor-induced immune suppression and activate antitumor immune responses. Although cytotoxic T lymphocytes (CTLs) play a pivotal role in this process, natural killer (NK) cells have also demonstrated remarkable tumor-killing abilities, given their ability to discriminate tumor cells from normal cells and […]

Driver mutations in GNAQ and GNA11 genes as potential targets for precision immunotherapy in uveal melanoma patients

21 de December de 2023/in Alemany’s & Piulats’s lab/by

Oncoimmunology. 2023 Oct 24;12(1):2261278. doi: 10.1080/2162402X.2023.2261278. eCollection 2023. ABSTRACT Uveal melanoma (UM) is the most common ocular malignancy in adults. Nearly 95% of UM patients carry the mutually exclusive mutations in the homologous genes GNAQ (amino acid change Q209L/Q209P) and GNA11 (aminoacid change Q209L). UM is located in an immunosuppressed organ and does not suffer […]

Novel combinatorial therapy of oncolytic adenovirus AdV5/3-D24-ICOSL-CD40L with anti PD-1 exhibits enhanced anti-cancer efficacy through promotion of intratumoral T-cell infiltration and modulation of tumour microenvironment in mesothelioma mouse model

6 de December de 2023/in Alemany’s & Piulats’s lab/by

Front Oncol. 2023 Nov 20;13:1259314. doi: 10.3389/fonc.2023.1259314. eCollection 2023. ABSTRACT INTRODUCTION: Malignant mesothelioma is a rare and aggressive form of cancer. Despite improvements in cancer treatment, there are still no curative treatment modalities for advanced stage of the malignancy. The aim of this study was to evaluate the anti-tumor efficacy of a novel combinatorial therapy […]

Overall Survival From Tebentafusp Versus Nivolumab Plus Ipilimumab in First Line Metastatic Uveal Melanoma: A Propensity Score Weighted Analysis

4 de December de 2023/in Alemany’s & Piulats’s lab/by

Ann Oncol. 2023 Dec 2:S0923-7534(23)05100-1. doi: 10.1016/j.annonc.2023.11.013. Online ahead of print. ABSTRACT BACKGROUND: Tebentafusp demonstrated a superior overall survival (OS) benefit (hazard ratio [HR] 0.51) compared to investigator’s choice (82% pembrolizumab) in a randomized, Phase 3 trial (IMCgp100-202; N=378) in untreated metastatic uveal melanoma (mUM). The 1-year OS rates for tebentafusp and pembrolizumab were 73% […]

Pembrolizumab and Enzalutamide in Patients with Abiraterone Acetate-Pretreated Metastatic Castration-Resistant Prostate Cancer: Cohort C of the Phase 1b/2 KEYNOTE-365 Study

8 de November de 2023/in Alemany’s & Piulats’s lab/by

Eur Urol Oncol. 2023 Nov 6:S2588-9311(23)00224-9. doi: 10.1016/j.euo.2023.10.008. Online ahead of print. ABSTRACT BACKGROUND: Limited responses have been observed in patients treated with enzalutamide after disease progression on abiraterone for metastatic castration-resistant prostate cancer (mCRPC), but androgen receptor signaling impacts T-cell function. OBJECTIVE: To evaluate the efficacy and safety of pembrolizumab plus enzalutamide in mCRPC. […]
